I am new to this and have read many of the posts in this forum but was unable to come up with an answer.
I am trying to define the brands of my devices, but the button is greyed out and in not acessible.
I first place the Neo in "use mode" by holding down the "mode" button for three seconds. Then the options appear. Only "use", "label", and "setup" are available.
What am I doing wrong?

You are probably pressing MODE while on the start up page. You need to go to a device page first. For example, from the start page, press TV and then you will be on TV page 1. Now press and hold MODE.
If you enter the MODE screen from the start page, you won't be able to do Macro, Brand, or Learn because these are device specific (i.e. apply only to the TV or only to the VCR, etc).

I had the same issue before. Those are the only options available at the "home" menu. If you select a device from the home menu then you also get Learn and Macro. So hit amp then hold mode for 3 seconds, that should work
